Abstract

Objective: Osteosarcoma is the most common primary bone cancer and, despite the advent of chemotherapy, the overall survival rate is low, especially when patients develop metastasis. Studies indicate the role of the CXCR4/CXCL12 axis in the development of metastases and in the reduction of the overall survival of patients. However, in vitro studies are needed to understand its role in the mechanisms of metastasis and chemoresistance in this tumor. The aim of this study was to evaluate the expression of the CXCR4 receptor and the secretion of its ligand, CXCL12, in primary osteosarcoma cells. We isolated two primary cell cultures, called OS1 and OS2, obtained from osteosarcoma samples from patients diagnosed at INTO. The osteosarcoma lineages MG-63 and SAOS-2 were used in the study Primary cells OS1 and OS2 exhibited morphology similar to that of the MG-63 line, in monolayer, and presented, respectively, mean duplication time of 75.70 and 29.97 hours. The sensitivity of osteosarcoma cells to the chemotherapy drugs doxorubicin (DOX), cisplatin (CIS) and methotrexate (MTX) were assessed by MTT assay and IC50 calculation. OS1 cells had the lowest IC50 values related to treatment with DOX, CIS, and MTX (concentrations of 0.40, 97.60 and 15.90 μM, respectively) and were, therefore, the most sensitive. On the other hand, OS2 cells had the highest IC50 values for DOX (97.20 μM) and for MTX (2231 μM). The concentration found for CIS (257.70 μM) was slightly lower than the higher one (309 μM - SAOS-2). The MG-63 strain was the second most resistant to DOX (4.86 μM) and MTX (465.20 μM) and the third most resistant to CIS (228.40 μM). The SAOS-2 strain was the most sensitive to MTX (2.05 μM), the second most sensitive to DOX (1.36 μM), but the most resistant to CIS (309 μM). The expression of CXCR4 mRNA in OS1 cells was significantly lower in OS2 cells and in relation to the MG-3 lineage, but there was no significant difference in relation to the SAOS-2 lineage. OS2 cells, on the other hand, showed CXCR4 mRNA expression similar to MG-63 and less expression in relation to OS-2 (p <0.01). In the immunofluorescence assay, weak CXCR4 labeling was observed in OS1 and in SAOS-2, whereas OS2 cells showed more intense labeling, similar to MG-63. By flow cytometry, it was found less than 1% of CXCR4 positive cells for OS1 while for OS2 the percentage ranged between 1.3% and 3%. Finally, the quantification of CXCL12 secretion by ELISA showed low autocrine secretion by OS1, OS2, MG-63 and SAOS-2 cells, with no statistic difference. Together, the data suggest that the expression profile of CXCR4 in primary osteosarcoma cells and in commercial lineages is heterogeneous and that the cells with higher expression of CXCR4 (OS2 and MG-63) show higher resistance to the in vitro action of chemotherapeutic drugs.
