Objective: The giant cell tumor of the bone is a benign primary neoplasm of bone that can travel with metastasis in 2% of cases. The treatment for this pathology consists mainly in the surgical removal of the tumor. Recent studies point to the CD90 surface protein as an important molecule in the oncogenesis process, but there are no reports in the literature investigating its role in GCT. In this context, understanding the role of this molecule in the natural history of GCT is important. The aim of this study is to investigate the expression of CD90 in this tumor and to explore its relation with cell proliferation and migration. CD90 expression was accessed in primary tumor samples from 23 cases (10 samples from patients with metastasis and / or relapse and 13 from patients without metastasis and / or relapse) and evaluated by immunohistochemistry. Subsequently, it was classified from zero (absent) to three (intense) considering the dispersion and the intensity of the marking in the histological section, with variability in the expression without difference between the groups (p = 0.595). Cellular expression of CD90 was evaluated in cells isolated from the tumor tissue of three patients by flow cytometry. In these 37.3% of the cells were CD90 +. Then, we performed the analysis of CD90 expression in primary cells and observed a percentage of 93% to 99% of CD90 + cells. In order to investigate its function in TCG, we evaluated the effect of silencing on the migration and proliferation of primary culture cells and as a result we obtained a significant reduction in expression of this molecule between days 7 and 12 after transfection (p <0.0001), With the return of the expression happening around the 14th day after electroporation. To evaluate the functionality of CD90 in TCG cells, cell migration and proliferation analysis assays were performed. In the SCR condition, a migration rate of 85.4% was observed, and in the condition that CD90 was silenced, there was a reduction in migration to 61.9%. The proliferative activity was analyzed by immunocytochemistry for Ki67. The mean number of Ki67 cells in the control, SCR and CD90 SiRNA conditions was 18.23 ± 3.3, 12.21 ± 3.8 and 15.82 ± 2.0, respectively (Value p = 0.412), indicating That silencing does not interfere with cell proliferation. Therefore, primary tumor tissue and GCT recurrence express CD90 (Thy-1), but unrelated to the occurrence of recurrence. In vitro, isolated cells also show expression of this molecule that is related to cell migration. | |
